Determination of protein concentration with two-dimensional (2D) Photon burst diagrams using microfluidic channel

Abstract

The determination of target protein concentration has been demonstrated by a 2D photon burst (photon counts vs. burst width) analysis technique and microfluidic channel device. Since this technique is based on counting individual molecule events, we can effectively estimate the concentration of the specific target molecule among other molecules in a sample solution in a simpler, faster and more accurate way comparing to the conventional ensemble measurement techniques. It is also possible to study protein-protein interaction with only one fluorescent label. This could find potential applications in clinical diagnostics and signal pathway study in the future.
