Abstract
Since Fidler established his method of orthotopic implantation, it has become indispensable for metastasis research. Three methods for orthotopic implantation of stomach cancer have been reported: Fidler's original method of injecting a cell suspension into the gastric wall; ligating tumor tissue to the serosa of stomach: and injecting minced tissue into the gastric wall. In this paper, we clarified the characteristics of each method and compared the technique, the similarity of patients and the metastatic rate. The second method is the easiest technique. The first method is most similar to patient. Because cells were injected into submucosa of stomach on the other hand, the second method involved serosa and the third method the submucosa and muscle layer. The rare of liver metastasis was much higher m using tumor tissue (second and third method than in using cell suspension first method). These results suggest that we should select the method of orthotopic implantation for the purpose of metastasis experiments.
