Separation of isolated Chinese hamster metaphase chromosomes into three size-groups

Abstract

A method is described for the isolation of metaphase Chinese hamster chromosomes and their separation into three size-groups. DNA extracted from fractionated chromosomes does not show heterogeneity in thermal denaturation temperature or buoyant density. The separation technique has been used to demonstrate a high rate of DNA synthesis in small hamster chromosomes during late S phase.
