Determination of lomefloxacin in human plasma by HPLC and its pharmacokinetics of intravenous infusion

Abstract

A rapid, sensitive and specific method for determination of lomefloxacin in human plasma by HPLC was proposed. The recovery from plasma was over 80% and detect limit 0.12 μmol · L-1. Pharmacokinetics of the drug after 1 h intravenous infusion of 200 mg lomefloxacin was studied. The plasma concentration vs time data were better described by an open two compartment model. The pharmacokinetic parameters were estimated as follows: T( 1/4 α), 0.38 ± 0.10 h; T( 1/4 β), 8.13 ± 2.46 h; V(c), 45.7 ± 9.6 L; Cl, 15.55 ± 1.75 L · h-1 and MRT, 7.89 ± 0.96 h.
